Stem cells and advanced therapies

Líneas de investigación

The Stem Cells and Cell Therapy laboratory of the IIS Biobizkaia focuses its activity on the potential of human stem cells and their derivatives (extracellular vesicles (EVs)) for the study of the etiopathogenesis of diseases and for the development of new innovative and effective therapeutic strategies, with special emphasis on the so-called advanced therapies.

The in vitro generation of disease models based on human stem cells provides us with physiological and reliable experimental models, especially for those diseases where animal models do not reflect the phenotype observed in patients. These human experimental models are a fundamental tool that has allowed us to study the molecular etiopathogenesis of lipodystrophies that cause premature aging and in which we are carrying out screening and drug repositioning projects for a rare genetically based disease characterized by high fragility. bone, such as osteogenesis imperfecta.

The most translational line of the laboratory focuses on the use of these cells as therapy, in fact, we have led a national and multicenter clinical trial (code: TERCELOI) of cell therapy applied to patients pediatric patients suffering from the rare genetically based disease, osteogenesis imperfecta (OI). TERCELOI has demonstrated the safety and therapeutic potential of 5 infusions of histocompatible mesenchymal stem cells in two pediatric patients with OI. This treatment resulted in a decrease in the number of fractures, and improvement in both bone parameters and the quality of life of the patients. The study of the mechanism of action revealed the existence of a functional pro-osteogenic paracrine response in these patients, as a consequence of cell therapy.

Currently, several of our lines of research focus mainly on using the knowledge acquired with said trial to develop highly effective and democratized advanced therapies, based on extracellular vesicles enriched in functional factors. These therapies aim to alleviate diseases that cause bone fragility such as osteogenesis imperfecta and the prevalent osteoporosis, as well as pathologies related to aging (both that caused by the rare lethal genetic disease Hutchinson–Gilford progeria, and physiological aging).
